Naguib et al. show that SUPT4H1, proposed to selectively transcribe repeat-containing DNA, is required for the homeostatic maintenance of total RNA levels in human cells. These findings highlight a paramount safety liability associated with targeting this molecule for the treatment of repeat-associated neurodegeneration.
