Civenni et al. demonstrate that the BET protein BRD4 regulates Mff transcription and mitochondrial fission, which is essential for the self-renewal, survival, and expansion of prostate cancer stem cells (CSCs). Pharmacological inhibition of BRD4 represents a promising therapeutic vulnerability in prostate cancer.
