CRISPR-READI: Efficient Generation of Knockin Mice by CRISPR RNP Electroporation and AAV Donor Infection (Cell Reports)
 


Generation of knockin mice harboring large insertions or sequence substitutions has relied primarily on costly and laborious microinjection-based CRISPR technology. Chen and Sun et al. developed the CRISPR-READI approach combining AAV-mediated donor delivery with Cas9/sgRNA RNP electroporation to engineer multi-kilobase modifications in the mouse genome with high efficiency and throughput.
