Substrate Affinity and Specificity of the ScSth1p Bromodomain Are Fine-Tuned for Versatile Histone Recognition (Structure)
 


Bromodomains recognize acetylated lysines in histones and other nuclear proteins. Blus et al. show that the ScSth1p bromodomain is fine-tuned for promiscuous histone tail recognition. Mutations of single residues in the bromodomain--identified by analyzing its crystal structure--unexpectedly caused significant increases in acetylated peptide binding affinity and selectivity, respectively.
