Spt5 promotes promoter-proximal pausing, promoter escape, mRNA processing, and selective transcription of the mutant huntingtin gene. Bahat et al. identified Spt5-Pol II small-molecule inhibitors that mimic Spt5 knockdown effects and uncouple its different functions. The inhibitors uncovered Spt5 regulatory roles in metabolism and 3´ end processing of histone genes.
