An SH3PX1-Dependent Endocytosis-Autophagy Network Restrains Intestinal Stem Cell Proliferation by Counteracting EGFR-ERK Signaling (Developmental Cell)
 


Zhang et al. show that multiple genes in the endocytosis-autophagy network regulate intestinal stem cell proliferation, gut homeostasis, and lifespan by restraining EGFR-RAS-ERK pathway activity. Mutations in genes in this network are enriched in microsatellite instable-high and KRAS-wildtype colorectal cancers, suggesting an alternate route to EGFR-RAS-RAF-ERK-driven tumorigenesis.
