Engineering Forward Genetics into Cultured Cancer Cells for Chemical Target Identification (Cell Chemical Biology)
 


Target identification of small molecules remains a barrier for drug discovery. Here, Povedano et al. establish a platform for forward genetic screening using CRISPR/Cas9 to engineer DNA mismatch repair deficiency in cancer cell lines and demonstrate the feasibility of this approach to identify the known targets of three anticancer toxins.
