Stress-Induced Cellular Clearance Is Mediated by the SNARE Protein ykt6 and Disrupted by ?-Synuclein (Neuron)

Cuddy et al. found that the SNARE protein ykt6 plays a crucial role in proteostasis and lysosomal function by enhancing hydrolase trafficking under stressful conditions. Parkinson`s disease ?-synuclein impedes ykt6, causing imbalanced proteostasis and self-propagating protein accumulation. Ykt6 can be therapeutically targeted by farnesyltransferase inhibitors that restore trafficking and lysosomal function.
