Thoma et al. characterize the mechanical unfolding pathway and structural stability of the transmembrane ? barrel assembly factor BamA. Exposed to stress, BamA unfolds single ? hairpins with its mechanical stability depending on the POTRA domain, the extracellular lid domain, and on the membrane composition.
